Determination of Hydroxypropyl Methylcellulose (HPMC) Viscosity
Viscosity is the most critical quality indicator for HPMC, directly reflecting its thickening and water-retention capabilities; it is a standard parameter measured during both production and application, typically using a rotational viscometer in industrial settings.
The primary testing equipment includes a rotational viscometer, a constant-temperature water bath, a beaker, and a stirring device. Standard testing involves preparing an HPMC aqueous solution with a mass fraction of 2% and maintaining the test temperature at 20 ± 0.1°C.
To prepare the sample, HPMC powder is dispersed in hot water (80–90°C) and stirred thoroughly to prevent clumping. The mixture is then cooled in an ice bath under continuous stirring to facilitate gradual dissolution. Once cooled, air bubbles and surface foam are removed, and water is added to adjust the solution to the specified concentration. The prepared solution is placed in a constant-temperature water bath for a sufficient period to ensure a uniform and stable temperature throughout.
After calibrating the viscometer and selecting the appropriate spindle and rotational speed, the spindle is immersed in the test solution, ensuring it does not touch the beaker walls or bottom. Once the instrument reading stabilizes, the viscosity value is recorded; 2–3 parallel measurements are performed, and the average is taken as the final result.
Several key factors influence the testing process. Temperature significantly affects viscosity—rising temperatures cause a marked decrease in viscosity—so temperature control must be strict. The solution must be free of air bubbles; incomplete dissolution or the presence of bubbles can cause reading fluctuations. As HPMC is a non-Newtonian fluid, the measured viscosity changes if the spindle or rotational speed is altered; therefore, test reports must specify the spindle model and speed settings to facilitate data comparison. Testing vessels and spindles must be kept clean to prevent residual impurities from interfering with the results.
